Total triglyceride differentiation assays are performed in a 96-well format using subcutaneous preadipocytes. Compounds are tested for their ability to increase triglyceride accumulation during the differentiation process compared to the positive control (complete differentiation medium).
Triglycerides are quantified using a coupled enzyme-colorimetric reaction and compared to a standard curve. A DMSO vehicle is a negative control, and PPARα agonists are positive controls. This service includes 10 customer-defined compound concentrations, each tested in triplicate.
The optimal sample for the service is a compound dissolved in DMSO, PBS, or culture medium.
