Altogen Labs provide complete RNAi gene silencing services, including validation of gene silencing - performed using real-time qRT-PCR (mRNA expression level) or Western blot (protein expression level).

Altogen Labs offer siRNA design, synthesis, purification and functional testing to measure percent (%) of mRNA knockdown using HeLa cell line (or cell line of choice - 70 cell lines available).

Service includes: Design, synthesis, HPLC purification, and annealing of 5 different siRNA molecules targeting gene of interest. siRNAs are tested for gene silencing potency. Two out of five siRNAs are finally selected (siRNAs are guaranteed to provide at least 80% mRNA expression knockdown).

Validation of gene silencing (RNAi) effect performed by real-time qRT-PCR (mRNA expression) or Western Blot (protein expression). Client provides the gene name and accession number of the target gene and we’ll do the rest.

Another option for client is to provide gene name and accession number of the target gene(s), mail the cells and primary antibodies for Western blotting analysis to Altogen Labs, and the lab team does the rest. Altogen’s shRNA knockdown service includes design, construction, and confirmation of the 3'-5' sequence the shRNA vectors for every target gene. Design and synthesis of the SYBR-labeled qPCR primers for quantitation of target gene expression, shRNA transfection into the cell line supplied by the customer, and targeted gene expression analysis by qPCR at 24, 48, and 72 hr post-transfection and Western blot at 48 and 72 hr after transfection are performed by company scientists. Altogen Labs guarantees 80% or more inhibition of target gene expression. Transfection controls includes the negative vector control with scrambled sequences (21 bp) and a GFP protein vector control for assessment of transfection efficiency.
