Altogen Labs provides RNAi reporter assay and RNAi cell line development services. 

RNAi stable cell lines expressing shRNA construct (to target gene of interest in long-term experiments ) are important gene expresion research tool. However, generation of RNAi stably-expressing cancer cell lines can be very laborious, expensive, and time-consuming. Altogen Labs offer generation of clonal stable knockdown cell lines based on client individual specifications. Our scientists bring years of extensive cell culture experience and expertise. Service includes:

a) Cloning: plasmid DNA encoding antibiotic-resistant gene, synthesis and cloning of shRNA construct targeting gene of interest into plasmid DNA, final plasmid DNA amplification, Maxi-prep purification.

b) Stable cell line generation: transfection of plasmid DNA (at least 20 ug should be provided by client) into a cell line of choice, drug selection of clonal cells, generation of stably-expressing population of cells, validation of shRNA construct expression and RNAi gene silencing by real-time qRT-PCR (mRNA expression) or Western Blot (protein expression).

Another option is generation of Tetracycline inducible RNAi stably-expressing cell lines.
