Liposome Encapsulation of small interfering RNA (siRNA) Molecules

Liposomes are composite structures made of phospholipids that can be made to encapsulate various types of nucleic acids within the aqueous core of the liposome.

Liposomes are used for the highly efficient delivery of cargo molecules (DNA, RNA, and protein) into cells both in vitro and in vivo. Altogen Labs offers encapsulation of negatively charged molecuIes (e.g., siRNAs, shRNAs, microRNAs, mRNAs, plasmid DNAs, and proteins) into either one of several standard liposome formulations (PC:Cholesterol and DSPC:Cationic Lipid:PEGylated Lipoid:Cholesterol) or a custom liposome formulation.

Sonication, liposome extrusion, and microfluidization (microfluidics) methods allow the generation of liposomes of a defined size (from 50 nm to 400 nm). The customer should provide at least 0.25 mg siRNA for liposome encapsulation. Liposome-encapsulated siRNAs are delivered to the customer.

Custom experimental design is available to provide in vivo siRNA protection (in the liposomal core), PEG-modifications to reduce the innate immune response, and low cytotoxicity using biodegradable lipids and polymers.

Altogen Labs previously developed a line of in vivo transfection products including PEG-liposomes and nanoparticle-, polymer- and lipid-based in vivo transfection systems for laboratory testing of pharmaceuticals in cells and animals (www.altogen.com/mirna.htm).

Once we know the details of your project, we can provide you an immediate price quote (contact e-mail: info@altogenlabs.com). Please note that experimental details will help us to provide an accurate quote and timeline estimate.
