Transient Transfection of siRNA, microRNA, mRNA, or plasmid DNA
Altogen Labs offers transient transfection of siRNA, microRNA, shRNA, and plasmid DNA into the cell line of choice. The standard service includes the optimization of transfection of DNA or RNA (at least 20 µg should be provided by the client) into the cell line of choice and final validation of construct expression by real-time qRT-PCR (i.e., mRNA expression) and/or Western blot (i.e., protein expression).
Gene Knockdown: Stable RNAi Cell Line Development
This service includes the generation of stable RNAi cell lines expressing shRNAs, targeting genes of interest. Such cell lines are important gene expression research tools that provide valuable information on gene function, pathway signaling, and long-term silencing effects. However, the generation of stable RNAi -expressing cancer cell lines is a long and complex process. Altogen Labs offers its expertise and extensive experience to develop stable knockdown cell lines. The client provides the gene ID as the target, and company scientists complete the project based on its requirements and specifications.
The standard service includes plasmid DNAs encoding antibiotic-resistant genes, synthesis and cloning of shRNA constructs targeting the genes of interest into plasmid DNAs, final plasmid DNA amplification, Maxi-prep purification, stable cell line generation, transfection of plasmid DNAs into the cell line of choice, drug selection of clonal cells, generation of stable cells, validation of shRNA construct expression, and RNAi gene silencing by real-time qRT-PCR (i.e., mRNA expression) or Western blot (i.e., protein expression). Two clonal cell lines (one cryovial each) are delivered to the customer.
